Overview

Heat shock protein 70 (HSP70)-derived peptide epitopes are specific amino acid sequences from the HSP70 chaperone family that serve as potent immunomodulatory targets in oncology. While HSP70 is primarily an intracellular protein, it is frequently expressed on the plasma membrane of various tumor cells, a phenomenon not observed in healthy tissues (Multhoff, 2007). These membrane-bound epitopes, particularly the 14-mer TKD peptide (TKDNNLLGRFELSG), act as recognition motifs for Natural Killer (NK) cells via the CD94/NKG2C receptor complex (Krause et al., 2004). Interaction with these epitopes triggers NK cell activation, migration, and the release of granzyme B, leading to the targeted destruction of malignant cells (Specht et al., 2015). In therapeutic development, these peptides are utilized as vaccines or in ex vivo cell therapy protocols to prime the patient's immune system against HSP70-positive tumors (Shevtsov et al., 2018). Consequently, these epitopes represent a unique class of tumor-specific antigens that facilitate the selective elimination of cancer cells while minimizing off-target effects on normal physiology.
