Endoplasmin, widely known as gp96 or GRP94, is the primary molecular chaperone residing in the lumen of the endoplasmic reticulum (ER) [1]. It belongs to the HSP90 family and is essential for the folding and maturation of critical signaling proteins, including integrins and Toll-like receptors [4]. In the context of immunotherapy, the 'gp96-Ig' construct is a bioengineered fusion protein that combines gp96 with the Fc region of IgG1, allowing it to be secreted from cells rather than retained in the ER [2]. This secreted gp96-Ig acts as a powerful vaccine adjuvant by capturing tumor-derived peptides and delivering them to antigen-presenting cells through the CD91 receptor [2, 3]. This process triggers a potent T-cell mediated immune response, making it a significant target for the development of personalized cancer vaccines and therapies for infectious diseases [3].
