Overview
Heat shock protein family A member 2 (HSPA2) is a molecular chaperone of the highly conserved HSP70 family. HSPA2 helps maintain protein quality control by facilitating correct protein folding, preventing aggregation, and mediating degradation of irreparably damaged proteins through both ubiquitin-proteasome and autophagy pathways[1][3]. It is highly expressed in sperm cells, where it controls multiple aspects of spermatogenesis and sperm–egg recognition, and is also found in epithelial cells, where it influences differentiation and immunological signaling[5][4]. HSPA2 expression is associated with cancer cells, male infertility, and skin disease, and its dysregulation can result in defective cell differentiation and altered immune response[1][4][5]. While direct drug targeting of HSPA2 is still under investigation, inhibitors of the HSP70 family may impact its function as part of broader therapeutic approaches in cancer and stress-related diseases[1][3][6].

Mechanism of action
Inhibition of ATPase activity or substrate binding may disrupt its chaperone functions, protein folding, and stress response. Indirect targeting can occur via proteasome inhibitors or general HSP70 family inhibitors. It also modulates downstream pathways including immune signaling and apoptosis.
