Overview

Heat shock protein family D member 1 (HSPD1, commonly called HSP60) is a mitochondrial chaperonin that plays an essential role in protein folding, refolding misfolded proteins, and preventing protein aggregation. HSPD1 is critical for maintaining proper mitochondrial function and contributes to cell survival during physiological and environmental stress. In cancer, HSPD1 is often upregulated, promoting tumor cell survival, proliferation, metastasis (by repressing E-cadherin and enhancing migration/invasion), and angiogenesis. Extracellular HSPD1 has immune-modulating activities within the tumor microenvironment. Pathogenic mutations in the HSPD1 gene are associated with neurodegenerative diseases and may contribute to cardiovascular and inflammatory disorders due to disrupted protein homeostasis
