Heat shock transcription factor 5 (HSF5) is a member of the heat shock factor family of transcription factors and is characterized by a winged-helix-turn-helix DNA-binding domain. Unlike canonical HSFs such as HSF1, HSF5 lacks specific heptad repeats required for trimerization. HSF5 is predominantly expressed in the testis, where it is critical for the progression of meiotic prophase in spermatocytes and the successful completion of spermatogenesis under non-stress conditions. Its disruption leads to meiotic arrest and male infertility. HSF5 binds unique DNA motifs, partly distinct from other HSFs, and is a regulator of genes involved in chromatin organization and the meiotic program. HSF5 has emerging relevance as a biomarker for testicular function and cancer prognosis[1][2][3][4][5][7].
