Overview

Hedgehog acyltransferase-like protein (HHATL) is a human protein coded by the HHATL gene, located on chromosome 3. It is part of the membrane-bound O-acyltransferase (MBOAT) superfamily. Unlike its better-characterized relative Hedgehog acyltransferase (HHAT), HHATL does not itself exhibit enzymatic activity in palmitoylating Hedgehog proteins. Instead, HHATL negatively regulates the N-terminal palmitoylation of Sonic Hedgehog (SHH) by interacting with HHAT, thereby acting as a modulator of the Hedgehog signaling pathway[3]. Additionally, HHATL has been shown to ameliorate endoplasmic reticulum stress via the activation of autophagy, through interactions with the protein LC3[4]. The physiological and pathological roles of HHATL remain under investigation, with no current confirmation as a direct therapeutic target or biomarker. Alternative names for HHATL include C3orf3, GUP1, MBOAT3, and other synonyms relevant to its sequence homology and proposed functions.
