Overview

The Hedgehog signaling pathway is a critical regulator of embryonic development and adult tissue homeostasis. It consists of several key molecular components including the secreted ligand Sonic hedgehog (SHH), its primary receptor Patched 1 (PTCH1), the signal transducer Smoothened (SMO), and downstream transcription factors such as the GLI family. Activation begins when SHH binds PTCH1 on target cells, relieving inhibition on SMO and triggering intracellular cascades that ultimately regulate gene expression via GLI transcription factors. Aberrant activation or mutation within this pathway is implicated in various cancers—including basal cell carcinoma—and developmental disorders. Several drugs targeting SMO are approved for cancer therapy; however, safety concerns exist due to the essential role of this pathway in normal tissue maintenance and embryogenesis[1][2].
