Overview

The Helicobacter pylori cell envelope is a complex, multi-layered structure that serves as the primary interface between the bacterium and the human gastric environment. It consists of an inner cytoplasmic membrane, a thin peptidoglycan layer, and an asymmetric outer membrane containing lipopolysaccharides (LPS) and a variety of outer membrane proteins (OMPs) (Kusters et al., 2006). These components are essential for maintaining structural integrity, protecting the bacterium from gastric acid, and mediating adhesion to the gastric epithelium via proteins like BabA and SabA (Salama et al., 2013). The cell envelope also plays a critical role in pathogenesis by housing the Type IV secretion system, which delivers virulence factors like CagA into host cells (Odenbreit, 2005). From a therapeutic perspective, the cell envelope is a major target; for instance, beta-lactam antibiotics like amoxicillin act by inhibiting the synthesis of the peptidoglycan layer, leading to osmotic lysis of the cell (Kao et al., 2016). However, the envelope's low permeability and the presence of efflux pumps present significant challenges for antibiotic efficacy and contribute to the development of drug resistance (Tegtmeyer et al., 2011).
