Overview

Helicobacter pylori enzymes and cell envelope components represent a diverse group of molecular targets essential for the survival, colonization, and pathogenesis of the bacterium within the acidic environment of the human stomach. The enzyme urease is perhaps the most critical, as it catalyzes the hydrolysis of urea into ammonia and carbon dioxide, effectively neutralizing local gastric acid to allow bacterial persistence (StatPearls, 2023). The cell envelope, characterized by a Gram-negative structure, includes a peptidoglycan layer and an outer membrane rich in phospholipids, lipopolysaccharides, and specialized adhesins like BabA and SabA that mediate attachment to gastric epithelial cells (PubMed, PMID: 11553312). Therapeutic strategies typically involve a combination of antibiotics that target these components, such as amoxicillin (inhibiting cell wall synthesis) and clarithromycin (inhibiting protein synthesis) (PubChem). Eradication of H. pylori through these targets is the standard of care for preventing peptic ulcer recurrence and reducing the incidence of gastric adenocarcinoma and MALT lymphoma (NIH, 2022). However, the rapid rise of antibiotic resistance in these specific enzymatic and structural targets poses a significant challenge to global health.
