Overview

Helicobacter pylori proteins and enzymes encompass the collective proteome of the H. pylori bacterium, including essential metabolic enzymes and virulence factors required for gastric colonization (StatPearls, 2023). A primary target is urease, which allows the pathogen to survive the highly acidic stomach environment by generating ammonia (PubMed, PMID: 21254514). Other critical proteins include the vacuolating cytotoxin A (VacA) and cytotoxin-associated gene A (CagA), which are instrumental in host cell damage and the development of gastric cancer (Amieva & Peek, 2016). Therapeutic intervention typically involves multi-drug "cocktails" including antibiotics like clarithromycin, which inhibits the 50S ribosomal subunit, and amoxicillin, which targets penicillin-binding proteins to disrupt cell wall synthesis (PubChem). The clinical management of H. pylori is increasingly complicated by mutations in targets like DNA gyrase (gyrA) and 23S rRNA, leading to widespread antibiotic resistance (WHO, 2017). This broad target category is central to the treatment of peptic ulcers and the prevention of gastric malignancies. Research continues into novel inhibitors of H. pylori-specific metabolic pathways, such as the shikimate pathway, to overcome existing resistance mechanisms (PubMed, PMID: 30254245).
