The Helper T cell type 2 to helper T cell type 1 switch refers to the process by which CD4+ T helper cells (Th2) committed to a type 2 immune response can be reprogrammed or redirected toward a type 1 (Th1) phenotype. This switch is significant in immunology because Th1 and Th2 cells orchestrate distinct immune responses: Th1 cells promote cellular immunity against intracellular pathogens, while Th2 cells support humoral immunity and are associated with allergic responses. The switch involves downregulation of GATA3/STAT6-driven programs and upregulation of T-bet/STAT4-driven programs under influence of environmental cues like increased levels of IFN-γ or therapeutic administration of cytokines such as IL-12.
