Overview

Hemagglutinin (HA) is the major surface glycoprotein of the H5N1 Influenza A virus, playing a critical role in viral pathogenesis by mediating binding to host cell sialic acid receptors and facilitating membrane fusion (UniProt, 2024). In the context of inactivated H5N1 vaccines, the HA protein serves as the primary immunogen that induces the host's immune system to produce neutralizing antibodies (NIH, 2023). These antibodies target the globular head of the HA protein, effectively preventing the virus from initiating an infection (CDC, 2023). Because H5N1 is a highly pathogenic avian influenza strain with significant pandemic potential, targeting HA through vaccination is a key public health strategy (WHO, 2024). Inactivated viral antigens provide a safe method to expose the immune system to the structural components of the virus, such as HA and neuraminidase, without the risk of viral replication (FDA, 2020).

Mechanism of action

Inactivated H5N1 viral antigens, specifically the hemagglutinin (HA) protein, stimulate the production of neutralizing antibodies that bind to the HA protein on the virus surface, thereby blocking viral attachment to host cell sialic acid receptors and preventing infection (FDA, 2020; CDC, 2023).
