The Hemagglutinin antigen of Influenza A virus H1N1 subtype is a homotrimeric glycoprotein on the viral surface, composed of subunits HA1 and HA2 derived from cleavage of the precursor HA0. Its globular head domain binds sialic acid receptors on host cells, initiating infection, while the stem domain mediates fusion of viral and host membranes in acidic endosomes[1][3][5][2]. Hemagglutinin is the principal antigen for influenza immunity: neutralizing antibodies (often elicited by vaccination) bind to regions on HA, blocking viral attachment or fusion. Its frequent antigenic shifts and drifts cause seasonal flu outbreaks and occasional pandemics. HA's structure and function make it a prime target for therapeutic antibodies and ongoing universal vaccine research[1][5][3][4][2].
Neutralizing antibodies block receptor binding or fusion activity of HA. Vaccines elicit immune responses against HA, preventing viral entry.
