The hemagglutinin (HA) stalk domain is a highly conserved region of the influenza A virus HA protein, particularly within phylogenetic group 1 subtypes. It is located proximal to the viral membrane and is structurally distinct from the immunodominant globular head domain. The HA stalk plays an essential role in membrane fusion during viral entry and is a target for broadly neutralizing antibodies.
Antibodies targeting the HA stalk can neutralize diverse strains within group 1 by inhibiting membrane fusion or interfering with other steps required for infection. Some antibodies may also act via ADCC/ADCP or CDC.
