Overview

Hematopoietic cell-specific Lyn substrate 1 (HCLS1, also known as HS1) is an intracellular, actin-binding adaptor protein primarily expressed in hematopoietic cells. It is a cytoskeletal regulator closely related to cortactin, functioning by promoting dynamic actin remodeling required for leukocyte migration, antigen receptor signaling, adhesion, and immune synapse formation. HCLS1 mediates downstream signaling from multiple immune receptors, including the B-cell receptor (BCR), T-cell receptor (TCR), and chemokine receptor CXCR4. Altered HCLS1 expression or phosphorylation state is associated with various pathologies, including leukemia and altered immune responses; its low expression in lung adenocarcinoma correlates with poor prognosis and reduced immune cell infiltration. Commonly used as a biomarker in immune-oncology research, HCLS1 is not currently a direct drug target but remains of significant research interest for its roles in immunity and cancer biology.[1][3][5]
