Hematopoietic stem cell (HSC) function refers to the fundamental biological processes of self-renewal and multi-lineage differentiation that maintain the blood and immune systems throughout an organism's life [NIH, 2016]. These cells reside in specialized bone marrow niches where they remain primarily quiescent but can rapidly proliferate and differentiate in response to physiological stressors like hemorrhage or infection [Nature Reviews Molecular Cell Biology, 2012]. Proper HSC function is vital for hematologic homeostasis, and its impairment is linked to various diseases, including bone marrow failure syndromes and hematologic malignancies such as leukemia [Hematology/Oncology Clinics, 2017]. While 'function' itself is not a singular molecular target like a receptor or enzyme, it is the primary physiological endpoint for numerous therapeutic interventions, such as those using G-CSF (Filgrastim) or Plerixafor to mobilize cells for transplantation [FDA Label, 2008]. Therapeutic strategies often focus on modulating signaling pathways or niche interactions to enhance HSC expansion or protect the stem cell pool from damage during intensive chemotherapy [Cell Stem Cell, 2014]. Understanding the molecular drivers of HSC function, such as the CXCR4/SDF-1 axis, allows for the development of drugs that can precisely control stem cell behavior for clinical benefit [Blood, 2015].
Pharmacological agents do not target 'function' directly as a molecule; instead, they modulate it by interacting with specific receptors (e.g., CXCR4, G-CSFR) or signaling pathways (e.g., TPO signaling) to trigger stem cell mobilization, proliferation, or protection [FDA Label, 2008; Nature Reviews Molecular Cell Biology, 2012].
