Overview

The hematopoietic tissue microenvironment (bone marrow niche) is a complex ecosystem of hematopoietic stem and progenitor cells, stromal and endothelial cells, extracellular matrix components, and soluble factors (such as cytokines and growth factors), all within the bone marrow. This specialized environment provides critical signals and structural support necessary for the self-renewal, proliferation, lineage commitment, and maturation of hematopoietic (blood-forming) stem cells into all blood cell types. The microenvironment comprises both cellular components (including mesenchymal stem cells, fibroblasts, osteoblasts, adipocytes, vascular endothelial cells, and mature hematopoietic cells) and non-cellular elements (collagen, fibronectin, laminin, glycosaminoglycans) that together maintain normal blood cell production and immune competence. Dysfunction of this microenvironment can contribute to diseases such as cancer, bone marrow failure, and age-related hematopoietic decline.
