Overview

Heme-binding sites are specialized structural pockets within hemoproteins that coordinate a heme prosthetic group, typically an iron-protoporphyrin IX complex. These sites are critical for a wide array of biological processes, including the transport of diatomic gases like oxygen in hemoglobin and myoglobin, electron transfer in the respiratory chain via cytochromes, and enzymatic catalysis in proteins like cytochrome P450 and catalases (Source: UniProt, "Hemoprotein"). In many hemoproteins, the iron atom within the heme serves as the active center for binding ligands or performing redox chemistry. From a therapeutic perspective, these sites are targeted by various agents; for instance, antimalarial drugs like chloroquine interfere with the sequestration of toxic heme into hemozoin within the malaria parasite (Source: PubMed, PMID: 15110095). Additionally, many toxins like cyanide and carbon monoxide exert their lethal effects by binding with high affinity to the heme-binding sites of cytochrome c oxidase and hemoglobin, respectively (Source: NIH, "Carbon Monoxide Poisoning").
