The heme biosynthetic pathway is a highly conserved, essential metabolic process responsible for the production of heme, a prosthetic group critical for hemoglobin, myoglobin, cytochromes, and other hemoproteins. The pathway consists of eight enzymatic steps that convert simple precursors into the complex tetrapyrrole structure of heme. Defects or deficiencies in any enzyme can lead to disorders known as porphyrias or sideroblastic anemias depending on which step is affected. Regulation primarily occurs at the first step via feedback inhibition by heme on ALA synthase.
