The hemoglobin synthesis pathway refers to the coordinated biochemical processes that produce hemoglobin, the oxygen-carrying protein in red blood cells. This process involves two main components: heme synthesis and globin synthesis. Heme synthesis occurs primarily in erythroid precursor cells and liver cells, involving condensation of glycine and succinyl-CoA, formation of porphobilinogen, synthesis of protoporphyrin IX, and insertion of iron. Globin chains are synthesized on ribosomes and assemble with heme to form functional hemoglobin. Disruptions at any step can cause diseases such as porphyrias, sideroblastic anemias, thalassemias, or sickle cell disease.
