Target intelligence / Profile preview

heparan sulfate-glucosamine 3-sulfotransferase 4 (HS3ST4)

Target
HS3ST4
Molecular classification
Enzyme, Sulfotransferase
01

Overview

Heparan sulfate-glucosamine 3-sulfotransferase 4 (HS3ST4) is an enzyme that catalyzes the transfer of a sulfate group to the 3-O position of glucosamine residues in heparan sulfate chains, generating specific sulfated motifs used in cellular signaling and as attachment sites for herpesviruses. Expression of HS3ST4 is linked to viral entry and pathogenicity in herpes simplex and varicella-zoster viruses, and sequence variants in its gene have been associated with susceptibility to herpes zoster and postherpetic neuralgia

02

Mechanism of action

Not therapeutically targeted by approved drugs; potential mechanisms involve inhibition of enzymatic sulfotransferase activity or blockade of HS modification to prevent viral entry

03

Biological functions

Glycosaminoglycan biosynthetic processSulfotransferase activityInvolved in heparan sulfate modificationPotential role in viral entry/pathogenesis
04

Disease associations

Herpes simplex infectionHerpes zosterPostherpetic neuralgiapossibly other HS-related pathologies
05

Safety considerations

Not established, as HS3ST4 is not currently a direct drug target; general concerns would relate to off-target disruption of glycosaminoglycan biology
06

Interacting drugs

None directly known or clinically established as of now (HS3ST4 is not a widely drugged target)
07

Biomarkers

Variants in HS3ST4 (e.g., rs12596324 SNP) are associated with herpes zoster/postherpetic neuralgia risk; may be explored as susceptibility biomarkers
