The Adenovirus serotype 5 (Ad5) fiber shaft contains a KKTK motif that binds to the heparin-sensitive receptor on dendritic cells and monocytes. This binding facilitates viral entry and triggers the maturation of dendritic cells and the release of pro-inflammatory cytokines (such as IL-6 and TNF-alpha), which are necessary for the induction of potent adaptive T-cell and B-cell immune responses but also cause systemic pyrogenicity (fever).
