Overview

Hepatic lipid metabolism-related proteins refer to a broad functional group of enzymes, transcription factors, and transport proteins that coordinate the synthesis, storage, and breakdown of lipids within the liver (NIH, 2020). This category includes critical lipogenic enzymes such as Fatty Acid Synthase (FASN) and Acetyl-CoA Carboxylase (ACC), as well as master transcriptional regulators like Sterol Regulatory Element-Binding Proteins (SREBPs) and Peroxisome Proliferator-Activated Receptors (PPARs) (MDPI, 2021; Food & Nutrition Research, 2020). These proteins maintain systemic lipid homeostasis by balancing de novo lipogenesis with fatty acid beta-oxidation and the assembly of very-low-density lipoproteins (VLDL) for secretion (NIH, 2020). Dysregulation of these proteins is a primary driver of metabolic disorders, most notably Non-Alcoholic Fatty Liver Disease (NAFLD) and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD), where an imbalance leads to excessive triglyceride accumulation and lipotoxicity (Hokudai, 2021). Pharmacological agents often target specific members of this group; for instance, statins inhibit HMG-CoA reductase to lower cholesterol, while fibrates activate PPAR-alpha to enhance fatty acid oxidation (PubMed). Emerging therapies like resmetirom target the thyroid hormone receptor beta to reduce hepatic fat content in patients with steatohepatitis (FDA). Because this term describes a collective set of proteins rather than a single molecular entity, it is classified as a category of targets rather than a specific therapeutic target.
