Overview

The hepatic VLDL assembly pathway consists of molecular and cellular events in hepatocytes that coordinate the synthesis, lipidation, and secretion of very low-density lipoprotein particles. The process begins with the translation and translocation of Apolipoprotein B-100 in the rough endoplasmic reticulum, lipidation by microsomal triglyceride transfer protein, formation of nascent VLDL, and subsequent maturation and secretion into the bloodstream. VLDL particles transport endogenously synthesized triglycerides and cholesterol esters from the liver to peripheral tissues. Dysregulation of this pathway is implicated in metabolic diseases, including fatty liver, hypertriglyceridemia, and atherosclerosis.
