The hepatic VLDL synthesis pathway is a series of biochemical processes in the liver that produce and secrete VLDL particles, rich in triglycerides. Niacin modulates this pathway, leading to decreased VLDL production, lower triglycerides and LDL cholesterol, and increased HDL cholesterol. It acts by inhibiting triglyceride synthesis, reducing free fatty acid delivery to the liver, and increasing apoB degradation.
Niacin inhibits hepatic diacylglycerol acyltransferase-2, reducing triglyceride synthesis and VLDL production. It also reduces free fatty acid delivery to the liver and accelerates intracellular apoB degradation.
