Overview

Hepatitis B viral RNA refers to the RNA molecules transcribed in infected hepatocytes from the covalently closed circular DNA (cccDNA) of hepatitis B virus. The most notable forms are the pregenomic RNA (pgRNA)—serving as the template for reverse transcription into viral DNA—and several messenger RNAs encoding viral proteins, including surface (HBsAg), core (HBcAg), polymerase, and HBx. The pgRNA contains highly structured elements such as the epsilon stem-loop, which acts as a signal for encapsidation and directly participates in the formation of new viral particles[2][3]. Post-transcriptional modifications of HBV RNA, such as N6-methyladenosine (m6A), regulate its stability, efficiency of packaging, and successful viral replication[2]. Therapeutic intervention can aim at disrupting pgRNA synthesis, stability, or function, making HBV RNA an important molecular target for antiviral research and clinical monitoring[1][2][4].
