Hepatitis B virus (HBV) covalently closed circular DNA (cccDNA)-derived transcripts are RNA molecules transcribed from the cccDNA minichromosome within infected hepatocytes. These transcripts, including pregenomic RNA (pgRNA), precore RNA, surface antigen RNAs, and X gene RNA, serve as templates for viral protein synthesis and are essential for HBV replication and persistence. They are key biomarkers for monitoring active infection and therapeutic response, and represent therapeutic targets for achieving functional cure.
HBV antivirals can reduce cccDNA transcription activity or inhibit reverse transcription of pgRNA.
