Overview
The Hepatitis B virus (HBV) genome is a partially double-stranded, circular DNA molecule approximately 3.2 kilobases (kb) in length. It contains four major overlapping open reading frames (ORFs): S, C, P (Pol), and X, encoding essential viral proteins. The genome replicates via an RNA intermediate and reverse transcription. The covalently closed circular DNA (cccDNA) form persists in infected hepatocytes, serving as a reservoir responsible for chronic infection and remains untargeted by current antiviral therapies.
