Overview

The hepatitis B virus (HBV) packaging reaction refers to the highly specific process by which the viral pregenomic RNA (pgRNA), together with the viral polymerase, is selectively encapsidated into newly forming nucleocapsids during HBV replication. This step is essential for subsequent reverse transcription and production of infectious virions. Specificity is conferred primarily through interaction between HBV polymerase and the epsilon (ε) signal on pgRNA. Core protein alone can bind various RNAs non-specifically unless guided by this complex formation. Multiple host chaperones facilitate proper folding/assembly during this process in vivo. Disruption or modulation at any step can impair virus production, offering potential antiviral strategies.
