The Hepatitis B virus (HBV) polymerase, also known as Pol or P protein, is a multifunctional enzyme essential for the replication of HBV. It converts the viral pre-genomic RNA (pgRNA) into partially double-stranded relaxed circular DNA (rcDNA) within newly formed viral capsids. It possesses reverse transcriptase and RNase H activity. The polymerase is a major target for antiviral drugs like entecavir and tenofovir. Resistance mutations can arise due to error-prone viral replication.
