The Hepatitis B virus (HBV) X region RNA transcript is a vital component of the HBV life cycle, encoding the multi-functional HBx protein. This transcript is essential for the transactivation of both viral and host genes, playing a pivotal role in initiating and maintaining viral replication by stimulating the activity of covalently closed circular DNA (cccDNA) (Source: PubMed, PMID: 26071642). Beyond replication, the X region RNA and its protein product are strongly associated with the pathogenesis of hepatocellular carcinoma (HCC) through the disruption of host cell signaling, DNA repair mechanisms, and apoptotic pathways (Source: NIH, NCBI Bookshelf). Because the HBV genome consists of overlapping open reading frames, the X region sequence is present in nearly all viral mRNA species, making it a highly efficient target for therapeutic silencing (Source: Journal of Hepatology, 2021).
