Overview

Hepatitis C virus non-structural protein 3 (NS3) is a 70 kDa multifunctional enzyme produced by cleavage of the HCV polyprotein. It has two main functional domains: an N-terminal serine protease (involved in processing the viral polyprotein into mature non-structural proteins) and a C-terminal helicase/nucleoside triphosphatase (critical for viral RNA replication). The protease domain requires a cofactor, NS4A, for full activity. NS3 is an essential component of the HCV replication complex and a validated target for several clinically used direct-acting antiviral drugs. Its proper function is necessary for the life cycle of the hepatitis C virus, making it a central therapeutic target in HCV infection management.
