Hepatitis C virus non-structural protein 5B (NS5B) is an RNA-dependent RNA polymerase (RdRp) essential for HCV genome replication. It catalyzes the synthesis of new viral RNA strands, making it a prime target for antiviral drugs. Its structure, resembling a right hand, contains a unique active site in the palm domain. NS5B interacts with other viral and host proteins, influencing replication efficiency. Inhibitors target either the active site (nucleotide analogs) or allosteric sites (non-nucleoside inhibitors) to disrupt its polymerase function.
