The Hepatitis C virus (HCV) NS3/4A protease is a membrane-associated, chymotrypsin-like serine protease essential for the viral life cycle. It cleaves the HCV polyprotein at multiple sites, generating mature viral proteins required for replication. It also disrupts host innate immunity by cleaving key signaling proteins like TRIF and MAVS. Due to its critical role, it is a major target for direct-acting antiviral drugs.
