The Hepatitis C virus (HCV) nonstructural protein 3 protease, commonly referred to as NS3 protease, is a multifunctional viral enzyme essential for HCV replication and maturation. The primary role of the NS3/NS4A complex is to cleave specific sites within the HCV polyprotein at junctions between nonstructural proteins: specifically at NS3/NS4A, NS4A/NS4B, NS4B/NS5A, and NS5A/NS5B sites. As an essential enzyme for HCV life cycle progression, the HCV NS3/NS4A protease has been established as one of the most attractive targets for anti-HCV drug development.
