The Hepatitis C virus NS4 protein refers to the nonstructural proteins NS4A and NS4B, which are produced from the viral polyprotein. Both play crucial roles in the HCV life cycle, particularly in viral replication and modulation of host cell processes. NS4A acts as a cofactor for the HCV serine protease activity of NS3 and impairs antigen presentation. NS4B is an integral membrane protein that induces dramatic rearrangements of intracellular membranes to form a "membranous web," which serves as a scaffold for assembling the viral replication complex and inhibits host antiviral responses. Both NS4A and NS4B are actively pursued as antiviral drug targets due to their essential roles.
