Hepatitis C virus nonstructural protein 4A (NS4A) is a small, multifunctional protein essential for the hepatitis C virus (HCV) life cycle. It is one of the nonstructural proteins encoded in the carboxy-terminal region of the HCV polyprotein and plays critical roles in viral replication, assembly, and immune evasion. It acts as a cofactor for the NS3 serine protease, anchors the NS3/NS4A complex to intracellular membranes, enhances helicase activity of NS3, regulates hyperphosphorylation of NS5A, and is involved in viral assembly and envelopment. Inhibitors targeting the NS3/NS4A protease have been developed as direct acting antivirals (DAAs), significantly improving treatment outcomes.
