Hepatitis C virus nonstructural protein 4B (NS4B) is a multifunctional, integral membrane protein essential for the replication and assembly of the hepatitis C virus (HCV). It plays a central role in viral RNA replication, organization of the replication complex, and modulation of host cell membranes. NS4B induces dramatic alterations in host cytoplasmic membranes to form a structure known as the "membranous web," which serves as a scaffold for assembling the HCV RNA replication complex. It also binds specifically to viral RNA and exhibits NTPase activity, contributing directly to viral particle assembly beyond its role in genome replication. Its unique functions make it an attractive target for antiviral drug development aimed at treating chronic hepatitis C infection.
