The Hepatitis C virus (HCV) nonstructural protein 5 (NS5) region encodes two distinct proteins: NS5A and NS5B. Both proteins are essential for viral replication and pathogenesis. NS5A is a multifunctional regulator involved in viral replication complex assembly and immune evasion. NS5B is an RNA-dependent RNA polymerase that catalyzes the synthesis of new HCV RNA strands. Both proteins are directly targeted by direct acting antivirals (DAAs).
NS5A inhibitors disrupt viral replication complex assembly and virion assembly; NS5B inhibitors block RNA-dependent RNA polymerase activity.
