The ORF2 protein is the major capsid protein of the hepatitis E virus (HEV). It is encoded by the second open reading frame (ORF2) of the HEV genome and plays a central role in viral structure, infectivity, immune evasion, and host interaction. It forms an icosahedral capsid and exists in various isoforms, including an infectious capsid-associated form, a glycosylated secreted form, and a cleaved secreted form. ORF2 interacts with host cell receptors such as heparan sulfate proteoglycans and ASGR1/ASGR2 to facilitate viral entry. It also modulates the host immune response. ORF2 contains neutralizing epitopes and is a promising candidate for subunit vaccine development and is also a diagnostic target in ELISA assays to detect acute versus chronic infections.
