Overview

Hepatocyte nuclear factor 4 alpha (HNF4α) mRNA is the messenger RNA transcript that encodes the HNF4α protein, a master transcriptional regulator primarily expressed in the liver, gut, and kidney (UniProt P41235). This mRNA serves as a critical template for the production of a protein that controls the expression of thousands of genes involved in nutrient metabolism, bile acid synthesis, and blood coagulation (PubMed: 30115707). In pathological states such as liver cirrhosis and hepatocellular carcinoma, HNF4α mRNA levels are often significantly downregulated, contributing to the loss of mature hepatocyte function and disease progression (PubMed: 24535413). Therapeutic interventions targeting this mRNA, such as small activating RNAs (saRNAs) like MTL-HNF4A, aim to restore HNF4α protein levels to re-establish normal cellular differentiation and metabolic activity (MiNA Therapeutics). Additionally, mutations affecting the stability or translation of HNF4α mRNA are linked to Maturity-Onset Diabetes of the Young type 1 (MODY1), highlighting its importance in pancreatic beta-cell function (NIH: NBK1522).
