Overview

Hepatoma-derived growth factor-related protein 2 (HDGFL2) is a member of the HDGF family that contains an N-terminal PWWP domain, enabling it to bind specifically to methylated histones such as tri-methylated lysines 36 and 79 of histone H3 and di- and tri-methylated lysine 20 of histone H4. It acts as an epigenetic regulator of myogenesis, associates with the BAF chromatin-remodeling complex, promotes DNA double-strand break repair through homologous recombination, and determines the site of HIV-1 integration independent of LEDGF/p75. HDGFL2 is implicated in cellular growth control, particularly through cyclin D1 expression, and diseases related to its dysfunction include neurodegenerative disorders such as frontotemporal dementia and amyotrophic lateral sclerosis. There are currently no drugs known to target HDGFL2, and no established biomarker or safety profile in a therapeutic context.
