The heptad repeat 2 (HR2) domain is a conserved region within the S2 subunit of the coronavirus spike (S) glycoprotein. The spike protein is a class I viral fusion protein responsible for mediating viral entry into host cells by facilitating membrane fusion. The HR2 domain, together with heptad repeat 1 (HR1), plays a critical role in this process. During virus-cell entry, conformational changes in the spike protein bring together the HR1 and HR2 domains to form a six-helix bundle structure. This structural rearrangement brings viral and cellular membranes into close proximity, enabling their fusion—a critical step for viral genome delivery into host cells. It is a target for antiviral strategies and vaccine design.
