Herpes simplex virus (HSV) DNA polymerase is a key enzyme required for the replication of the viral genome during productive infection of HSV-1 and HSV-2. It is encoded by the UL30 gene and forms a holoenzyme with the UL42 processivity factor. The enzyme possesses DNA polymerase, 3'-5' exonuclease (proofreading), and ribonuclease H activities. It is the primary target for antiviral drugs like acyclovir, but resistance can develop through mutations in UL30 or viral kinases.
