Herpes simplex virus envelope glycoprotein D (gD) is an essential structural component of the herpes simplex virus (HSV) envelope. It mediates viral entry into host cells by binding to specific cellular receptors (Nectin-1, HVEM, Nectin-2, and modified heparan sulfate), thereby triggering the fusion of the viral envelope with host cell membranes. It is indispensable for HSV infectivity and a key target for antiviral drug development and vaccine design. The ectodomain undergoes conformational changes upon receptor engagement, initiating downstream events for membrane fusion. The cytoplasmic domain is involved in replication, envelopment, and spread.
Targeting gD prevents viral entry into host cells by blocking receptor binding and subsequent membrane fusion.
