The Herpes simplex virus type 1 (HSV-1) genome is a linear, double-stranded DNA molecule approximately 152,000 base pairs (bp) in length. It consists of two unique regions (UL and US) each flanked by inverted repeats. The genome encodes about 75 proteins and forms multiple isomeric forms due to recombination. During latency, the genome circularizes, and only a few RNAs are transcribed. It encodes determinants for drug resistance, virulence factors, and is a target for antiviral therapies including gene-editing approaches.
