Herpes simplex virus type 1 (HSV-1)-infected tumor cell lysis is the process by which HSV-1, particularly engineered or oncolytic strains, infects and destroys cancer cells. This involves direct oncolysis through viral replication within tumor cells and subsequent cell death, as well as immune activation via the release of tumor antigens and stimulation of both innate and adaptive immune responses. Engineered oncolytic HSV-1 vectors are designed for selective replication in malignant cells, often through the use of tumor-specific promoters or modifications that reduce neurovirulence while preserving oncolytic activity.
