Herpes simplex virus type 1 ribonucleotide reductase large subunit (HSV-1 RR1) is a 136,000–140,000 Da viral polypeptide forming part of the ribonucleotide reductase enzyme. Along with the small subunit (HSV-1 RR2), it catalyzes the reduction of ribonucleotides to deoxyribonucleotides, supplying essential precursors for viral DNA synthesis. HSV-1 RR1 possesses unique structural features, including a distinct N-terminal domain, and is indispensable for viral replication in non-dividing cells. Its absence impairs viral neurovirulence. Drugs targeting this enzyme have been developed to selectively inhibit viral replication with minimal impact on host enzymes.
