Herpesvirus DNA polymerase is a key viral enzyme essential for the replication of herpesvirus genomes. It is a primary target for antiviral drugs used to treat infections caused by human herpesviruses, including herpes simplex virus (HSV) and human cytomegalovirus (HCMV). The enzyme catalyzes viral DNA synthesis, possesses proofreading activity, and functions within a holoenzyme complex. It is inhibited by nucleoside analogs and non-nucleoside inhibitors. Drug resistance can develop through mutations in the polymerase gene. Herpesvirus DNA polymerase remains one of the most validated antiviral targets, and ongoing research aims to develop new classes of inhibitors that overcome resistance mechanisms.
