Overview

Heterogeneous nuclear ribonucleoprotein L-like (HNRNPLL) is an RNA-binding protein involved in the regulation of alternative splicing and mRNA stability, particularly in lymphocytes[1][2][3][4][5]. It serves as a master regulator of activation-induced alternative splicing in T cells, notably modulating the splicing of CD45, which is crucial for T-cell development and activation[1][3][4][5][7]. In B cells, HNRNPLL is induced during differentiation into plasma cells, where it shapes the transcriptome by regulating exon inclusion, mRNA stability, and the downregulation of specific transcription factors such as Bcl6[2][4]. Its activity is essential for the terminal differentiation of plasma cells and optimal immunoglobulin production[2][4]. HNRNPLL functions by binding preferentially to CA-dinucleotide repeats in pre-mRNA introns and 3′ untranslated regions, acting in a context-dependent manner to influence exon inclusion/exclusion or stabilize mRNA[2]. Disruption of HNRNPLL can impair immune cell function and differentiation, but it is not an established therapeutic drug target[1][2][3][4][5][7][8].
